Uzun meslek yaşantısı süresince 200'ün üzerinde filmde, çoğunlukla psikopat kötü adamları canlandırdı. Adeta üzerine yapışmış olan bu kötü adam rollerinden en çok bilinenleri James Bond ve Cadılar Bayramı (Halloween) film serilerinde canlandırdığı karakterlerdir.

İngiliz Şövalyelik Nişanı'nı II. Dünya Savaşı sırasında Kraliyet Hava Kuvvetleri'nde bombardıman uçağı telsizcisi olarak gösterdiği yararlıklardan dolayı almıştır. 1944 yılında çıktıkları bombalama görevlerinden biri sırasında uçağı vurularak düşürülmüş, Donald Pleasence Almanlara esir düşmüştü. Esir tutulduğu kampta tiyatro oyunlarında rol almayı sürdürmüştü.

2 Şubat 1995 tarihinde Fransa'da geçirdiği bir kalp kapağı değişimi ameliyatının sonrasında ameliyatın komplikasyonları nedeniyle hayatını kaybetti. Öldüğünde 75 yaşındaydı. Cenazesi yakılmıştır.
